H₂N-PEG₃-OH | CAS: 6338-55-2 | Sinopeg April 2,2026.
1. Definition and Chemical Structure
Basic Information: It is a heterobifunctional PEG derivative, meaning it has two different reactive groups at each end. The "3" indicates three repeating ethylene glycol units.

Chemical Information:

CAS Number: 6338-55-2

IUPAC Name: 2-[2-(2-Aminoethoxy)ethoxy]ethanol

Molecular Formula: C₆H₁₅NO₃

Molecular Weight: Approx. 149.19 g/mol

Structural Formula: H₂N—CH₂CH₂—(OCH₂CH₂)₂—OH

2. Physicochemical Properties
Physical Form: Typically a colorless to light yellow transparent liquid, or a white/off-white solid at room temperature.

Solubility: Possesses both amino and hydroxyl groups, along with the PEG chain, making it soluble in water as well as in common organic solvents such as DMF, DMSO, and ethanol.

Chemical Properties:

Dual Reactivity: This is the core feature. The terminal amino group (-NH₂) is highly reactive, easily reacting with carboxyl groups (-COOH) or active esters (e.g., NHS ester) to form amide bonds. The hydroxyl group (-OH) at the other end can be used for esterification reactions.

Flexibility and Hydrophilicity: The central PEG chain provides good flexibility and water solubility, helping to improve the hydrophilic properties of the molecules it modifies.

3. Primary Application Areas
Drug Discovery & PROTACs: Serves as a linker in PROTACs (Proteolysis-Targeting Chimeras) and ADCs (Antibody-Drug Conjugates) to connect target protein ligands with E3 ligase ligands, precisely controlling the distance between molecules.

Bioconjugation: Utilizes the high reactivity of the amino group to attach drug molecules, fluorescent dyes, or targeting ligands to peptides, proteins, or antibodies for labeling, detection, or delivery purposes.

Materials & Surface Modification: Can be used to modify nanoparticles (e.g., magnetic nanoparticles, quantum dots) or medical device surfaces, introducing PEG chains to reduce non-specific protein adsorption and improve biocompatibility.

Hydrogels & Polymer Synthesis: Acts as a crosslinker or monomer in the construction of three-dimensional hydrogel networks, widely used in tissue engineering and drug delivery research.
